Desi Source Optimization

DESI source optimization is the systematic adjustment of desorption electrospray ionization parameters to improve the detection of molecules from biological surfaces and samples. The process tunes factors such as spray voltage, solvent composition, flow rate, emitter-to-surface distance, and spray angle, which control analyte desorption, ion formation, and transfer into the mass spectrometer. In bioengineering, optimized DESI sources support ambient mass spectrometry and chemical imaging of tissues, biomaterials, and engineered biological systems with minimal sample preparation. Careful optimization can increase signal intensity, spatial resolution, reproducibility, and molecular coverage, strengthening analyses of metabolism, disease-associated changes, and biomaterial performance.
