Glioma Sphere Culture

Glioma sphere culture is a three-dimensional in vitro technique for growing glioma cells as free-floating spheres, providing a model of tumor organization and cellular heterogeneity. Researchers typically dissociate glioma tissue or established cell lines and culture the cells under serum-free, nonadherent conditions supplemented with growth factors such as epidermal growth factor and basic fibroblast growth factor; sphere formation reflects the survival and expansion of cells with self-renewal capacity. In bioengineering, this system supports studies of glioma stem-like cells, differentiation, invasion, and treatment response, while enabling controlled drug screening and the development of more physiologically relevant tumor models.

Protocol for propagation of dissociated high grade glioma surgical specimens in serum-free neurosphere medium to select for cells with cancer stem cell phenotype. For specimens that fail to grow as neurospheres, an alternative protocol is suggested. A paraffin embedding technique for maintaining the 3D neurosphere architecture for immunocytochemistry is described.
