Hek293 Cells

HEK293 cells are an immortalized human cell line widely used in biomedical research and bioengineering because they grow readily in culture and efficiently express introduced genes. Derived from human embryonic kidney cells transformed with adenovirus 5 DNA, they support high levels of transfection, allowing researchers to produce recombinant proteins, study gene function, and test engineered genetic systems. In bioengineering, HEK293 cells serve as versatile platforms for evaluating molecular designs, manufacturing therapeutic proteins, and producing viral vectors for gene delivery. Their consistent growth and adaptable culture conditions make them valuable for both basic studies and development of biotechnology products.

Laboratory-scale production of eukaryotic proteins with appropriate post-translational modification represents a significant barrier. Here is a robust protocol with rapid establishment and turnaround for protein expression using a mammalian expression system. This system supports selective amino acid, selective labeling of proteins and small molecule modulators of glycan composition.

The molecular mechanisms that co-ordinate the formation of inhibitory GABAergic synapses during ontogeny are largely unknown. To study these processes,we have developed a co-culture model system which incorporates embryonic medium spiny GABAergic neurons cultured together with stably transfected human embryonic kidney 293 (HEK293) cells expressing functional GABAA receptors.

Small ubiquitin-related modifier (SUMO) family proteins are conjugated to the lysine residues of target proteins to regulate various cellular processes. This paper describes a protocol for the detection of retinoblastoma (Rb) protein SUMOylation under endogenous and exogenous conditions in human cells.
