Hepatocyte Isolation

Hepatocyte isolation is the process of separating liver parenchymal cells from tissue to obtain viable, functional cells for research and bioengineering. Typically, the liver is perfused with enzyme-containing solutions, such as collagenase, which disrupt the extracellular matrix and releases hepatocytes; gentle mechanical processing and centrifugation then enrich the cell fraction while preserving membrane integrity and metabolic activity. Isolated hepatocytes support studies of liver physiology, drug metabolism, toxicity, and disease, and provide building blocks for engineered tissues, organ-on-chip systems, and regenerative medicine. Cell yield, viability, and function depend on tissue handling, perfusion conditions, enzyme exposure, and culture environment.

Isolation of Human Hepatocytes by a Two-step Collagenase Perfusion Procedure

A modified two-step collagenase perfusion procedure for isolation of human hepatocytes is described. This method can also be applied to other mammalian livers. The isolated hepatocytes are available in high yield and viability, making them a suitable model for scientific research in areas such as liver regeneration, pharmacokinetics and toxicology.
