Hplc Gc-ms Analysis

HPLC-GC-MS analysis combines high-performance liquid chromatography, gas chromatography, and mass spectrometry to separate, identify, and quantify chemical components in complex samples. HPLC separates dissolved compounds according to interactions with a stationary phase and a flowing liquid, while GC vaporizes suitable analytes and resolves them in a capillary column; mass spectrometry then ionizes the separated molecules and measures their mass-to-charge ratios to generate identifying spectra. In bioengineering, these complementary methods characterize metabolites, pharmaceuticals, biomaterials, and bioprocess products. The resulting compositional and quantitative data support quality control, pathway analysis, process optimization, and evaluation of engineered biological systems.

The sample is either a gas or a liquid that is vaporized in the injection port. Typically, the compounds analyzed are less than 1,000 Da, because it is difficult to vaporize larger compounds. GC is popular for environmental monitoring and industrial applications because it is very reliable and can be run nearly continuously.

Here, we describe in great detail an established and robust protocol for the extraction of glucosinolates from ground plant materials. After an on-column sulfatase treatment of the extracts, the desulfoglucosinolates are eluted and analyzed by high-pressure liquid chromatography.

This study developed a high-performance liquid chromatography coupled with triple quadrupole mass spectrometry (HPLC-QQQ-MS) method to identify and quantify tropane alkaloids (hyoscyamine, anisodamine, scopolamine) in Anisodus tanguticus. The method proved rapid, accurate, and reliable for high-throughput analysis of medicinal plants.
