Microscope Hardware Setup

Microscope hardware setup is the selection, arrangement, and alignment of optical, mechanical, and electronic components needed to form reliable images of biological samples. Light from an illumination source is directed through or onto the specimen, while lenses collect and magnify transmitted, reflected, or fluorescent signals before a camera or eyepiece converts them into an observable image. In bioengineering, careful setup includes stable mounting, appropriate objectives, calibrated focus and illumination, and compatible image-acquisition hardware. These choices affect resolution, contrast, field of view, and measurement accuracy, supporting cell imaging, biomaterials characterization, microfluidic observation, and quantitative analysis.

A Low Cost Setup for Behavioral Audiometry in Rodents

A fast and inexpensive method for the behavioral determination of hearing parameters like hearing thresholds, hearing impairments or phantom perceptions (subjective tinnitus) is described. It uses pre-pulse inhibition of the acoustic startle response and can be easily implemented in a personal computer using a programmable AD/DA-converter and a piezo sensor.

Multiplexed ion beam imaging (MIBI) is often used to image tissue microarrays and tiled, contiguous tissue areas, but current software for setting up these experiments is cumbersome. The tile/SED/array interface is an intuitive, interactive graphical tool developed to dramatically simplify and accelerate MIBI run setup.

The system described herein employs a traditional optical trap as well as an independent holographic optical trapping line, capable of creating and manipulating multiple traps. This allows for the creation of complex geometric arrangements of refractive particles while also permitting simultaneous high-speed, high-resolution measurements of the activity of biological enzymes.

A wide-field Fourier-transform microscope, based on a compact and ultra-stable birefringent interferometer, allows the parallel acquisition of spectra for all pixels of a 2D detector. The time-domain approach enables the disentanglement of photoluminescence and Raman signals, and allows rapid Raman mapping (~5 ms/pixel) with ~1-µm spatial and 23-cm-1 spectral resolution.
