Nitrate Accumulation

Nitrate accumulation is the buildup of nitrate ions in biological tissues, soils, or engineered systems when inputs exceed uptake, assimilation, or removal. In plants, roots absorb nitrate through membrane transporters, and nitrate reductase converts it to nitrite before further assimilation into ammonium and amino acids; limited light, nutrient imbalance, or rapid nitrogen supply can disrupt this flow and increase storage. Bioengineering uses nitrate accumulation as a measurable trait in crop improvement, controlled-environment cultivation, and microbial or wastewater treatment systems. Monitoring it helps optimize nitrogen use, reduce environmental losses, and assess system safety and performance.

This video demonstrates an assay to measure the accumulation of a fluorescent antibiotic probe in bacterial cells. Exposure to an efflux pump inhibitor disrupts the transmembrane proton gradient in bacterial cells, upon which a fluorophore-conjugated antibiotic is added. Inhibition of efflux pumps due to the disrupted proton gradient causes accumulation of the antibiotic inside the bacteria. Bacterial cells are lysed, and the fluorescence intensity is measured to confirm antibiotic accumulation.

We present protocols for three different methods for the homogenization of four different muscle groups of rat skeletal muscle tissue to measure and compare the levels of nitrate and nitrite. Furthermore, we compare different sample weights to investigate whether tissue sample size affects the results of homogenization.
