Omental Tissue Injection

Omental tissue injection is a procedure that delivers cells, biomaterials, or therapeutic agents directly into the omentum, a vascularized fold of fatty tissue in the abdominal cavity. In bioengineering studies, a needle places the engineered payload within omental tissue, where local confinement, tissue contact, and blood supply can influence retention, integration, and biological activity. This approach supports evaluation of tissue-engineered constructs, cell-based therapies, drug delivery systems, and implantable materials in a physiologically relevant environment. By linking controlled fabrication with a living tissue site, omental injection helps researchers assess biocompatibility, vascularization, immune responses, and therapeutic performance before advancing regenerative strategies.

Here, we present a protocol for in vivo validation of hydrogel-based cell therapy, illustrated by the example of islet transplantation. h-Omental Matrix Islet filliNG (hOMING) implantation allows implantation of a cell-hydrogel mixture between the omental layers, near to blood vessels, to maximize engraftment in a proper metabolic environment.

The differentiation of white and beige adipocytes from adipose tissue vascular progenitors bears potential for metabolic improvement in obesity. We describe protocols for a CD34+CD31+ endothelial cell isolation from human fat and for a subsequent in vitro expansion and differentiation into white and beige adipocytes. Several downstream applications are discussed.
