Oxygen Barrier

An oxygen barrier is a material or layer that restricts the movement of oxygen, helping control gas exposure in biological, medical, and engineered systems. Its performance depends on oxygen permeability, solubility, diffusion through the material, and factors such as thickness, temperature, and concentration gradients. In bioengineering, oxygen barriers can regulate oxygen delivery around cells, tissues, implants, and biomaterials, while also limiting oxidative damage or unwanted reactions. Designing these barriers supports stable cell culture conditions, improves the preservation of oxygen-sensitive products, and enables better control of engineered tissue environments and device performance.

Ischemia-Reperfusion (IR) injury is associated with a high rate of morbidity and mortality. The goal of the in vitro model of oxygen-glucose deprivation and reoxygenation (OGD-R) described here is to assess the effects of ischemia reperfusion injury on a variety of cells, particularly in blood-brain barrier (BBB) endothelial cells.

Cerebral Blood Oxygenation Measurement Based on Oxygen-dependent Quenching of Phosphorescence

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

The aim of the protocol is to present an optimized procedure for the establishment of an in vitro blood-brain barrier (BBB) model based on primary porcine brain endothelial cells (pBECs).
