Spheroid Encapsulation

Spheroid encapsulation is the process of surrounding three-dimensional cell aggregates with a protective, often hydrogel-based, biomaterial matrix to control their local environment. The matrix immobilizes the spheroid while permitting the diffusion of nutrients, oxygen, signaling molecules, and waste, with material properties and capsule size influencing cell survival and function. In bioengineering, encapsulated spheroids support tissue engineering, drug screening, disease modeling, and studies of cell-cell interactions under physiologically relevant conditions. This approach can improve the handling and reproducibility of spheroid cultures while providing a platform for designing engineered tissues and investigating how cells respond to defined biochemical and mechanical cues.

Here, we present a protocol that enables fast, robust, and cheap fabrication of tumor spheroids followed by hydrogel encapsulation. It is widely applicable as it does not require specialized equipment. It would be particularly useful for exploring spheroid-matrix interactions and building in vitro tissue physiology or pathology models.

A method for photo-encapsulation of cells in a crosslinked PEG hydrogel is described. Hypoxic signaling within encapsulated murine insulinoma (MIN6) aggregates is tracked using a fluorescent marker system. This system allows serial examination of cells within a hydrogel scaffold and correlation of hypoxic signaling with changes in cell phenotype.

We present protocols for the 3-dimensional (3D) encapsulation of cells within synthetic hydrogels. The encapsulation procedure is outlined for two commonly used methods of crosslinking (michael-type addition and light-initiated free radical mechanisms), as well as a number of techniques for assessing encapsulated cell behavior.
