Voltage Calcium Imaging

Voltage calcium imaging is an optical approach for monitoring electrical activity and intracellular calcium dynamics in living cells, providing complementary views of how excitable cells signal. Voltage-sensitive indicators report rapid changes in membrane potential through fluorescence, while calcium indicators change fluorescence when they bind calcium ions released during or following electrical excitation. Bioengineers use these measurements to study neurons, cardiomyocytes, and engineered tissues, assess cellular connectivity and function, and evaluate disease models or therapeutic interventions. Combining voltage and calcium signals can distinguish fast electrical events from slower downstream calcium responses, supporting the design of biosensors, neural interfaces, and quantitative platforms for functional cell analysis.

This article describes the technique used to perform dual channel optical mapping in cultured HL-1 atrial cell monolayers. This unique protocol allows the simultaneous visualization of both calcium (Ca) and voltage (Vm) activity in the same area for the detailed detection and analysis of electrophysiological properties of culture monolayers.
