Whole Blood Assay

A whole blood assay is an analytical test performed directly on anticoagulated blood to measure cellular, biochemical, or immune responses while preserving interactions among blood cells, plasma proteins, and soluble factors. In a typical workflow, a blood sample is incubated with a stimulus, therapeutic compound, or engineered material, and the resulting changes are quantified through methods such as flow cytometry, immunoassays, or hematological analysis. These assays provide physiologically relevant data for evaluating inflammation, coagulation, immune activation, and blood compatibility. In bioengineering, they support the development of drug-delivery systems, biomaterials, medical devices, and diagnostic technologies with improved safety and performance.

Rapid Point-of-Care Assay of Enoxaparin Anticoagulant Efficacy in Whole Blood

Demonstration of a rapid quantitative assay of the inhibition of blood coagulation by the low-molecular-weight-heparin, enoxaparin. The contribution of enoxaparin is assessed by removing its influence through digestion with heparinase. A fuller description of the assay is detailed in our published paper.1 The assay still requires clinical confirmation.

In this report, we demonstrate the staining and analysis steps of a phenotyping assay performed on fresh whole blood to enumerate major innate and adaptive leukocyte populations. We emphasize considerations for performing these procedures in the context of a multicenter clinical trial.

Assaying Blood Cell Populations of the Drosophila melanogaster Larva

Drosophila blood cells, or hemocytes, cycle between resident sites and circulation. In the larva, resident (sessile) hemocytes localize to inductive microenvironments, the Hematopoietic Pockets, while circulating hemocytes move freely in the hemolymph. The goal of this protocol is the standardized isolation and quantification of these two, behaviorally distinct but interchanging, hemocyte populations.

The Micronucleus Assay on Cryopreserved Whole Blood

Here we present an optimized protocol for the cytokinesis-block micronucleus assay on cryopreserved whole blood samples. This optimized method of cryopreservation of whole blood for micronucleus analysis is a reliable technique for large-scale sampling and multi-center studies and can be used for other blood-related assays as well.
