Beta-lactamase Biosensor

A beta-lactamase biosensor is an analytical device that detects beta-lactamase activity, an important marker of bacterial resistance to beta-lactam antibiotics. The sensor uses a beta-lactam substrate that is hydrolyzed by the enzyme, converting the chemical reaction into a measurable signal such as a change in color, fluorescence, electrical response, or another physical property. By linking enzyme activity to signal intensity, these biosensors can support rapid screening of resistant bacteria, antibiotic susceptibility assessment, and monitoring of beta-lactamase contamination in biological or environmental samples. Their ability to provide sensitive, targeted measurements makes them valuable tools in microbiology, clinical diagnostics, and antibiotic-resistance research.

Effector translocation into host cells via a type III secretion system is a common virulence strategy among gram-negative bacteria. A beta-lactamase effector fusion based assay for quantitative analysis of translocation was applied. In Yersinia infected cells, conversion of a FRET reporter by the beta-lactamase is monitored using laser scanning microscopy.

Lytic phage biosensors and antibody beads are able to discriminate between methicillin resistant (MRSA) and sensitive staphylococcus bacteria. The phages were immobilized by a Langmuir-Blodgett method onto a surface of a quartz crystal microbalance sensor and worked as broad range staphylococcus probes. Antibody beads recognize MRSA.
