Total Protein Normalization

Total protein normalization is a quantitative strategy that expresses a protein signal relative to the total protein present in the analyzed sample, helping distinguish biological differences from variation in sample loading. In techniques such as Western blotting, researchers measure total lane protein using a stain or imaging-based signal, then divide the target-protein measurement by that value before comparing samples. This approach can reduce errors caused by unequal loading or transfer and may provide a broader reference than normalization to a single housekeeping protein. It supports more reliable protein-expression analysis in cell biology, disease research, and molecular biology experiments.
