Alpha Helix Domain

An alpha helix domain is a protein structural domain built primarily from alpha helices, a common arrangement that stabilizes proteins and helps determine how they interact with other molecules. In an alpha helix, a backbone carbonyl oxygen forms a hydrogen bond with the amide hydrogen four residues ahead, while side chains project outward and pack with neighboring helices or ligands. These domains support molecular recognition, DNA binding, membrane association, and assembly of multiprotein complexes; structural biology methods such as X-ray crystallography, nuclear magnetic resonance, and cryo-electron microscopy reveal how sequence changes alter folding, stability, and biological function.

Transmembrane Domain Oligomerization Propensity determined by ToxR Assay

An efficient procedure to assess the oligomerization propensity of single-pass transmembrane domains (TMDs) is described. Chimeric proteins consisting of the TMD fused to ToxR are expressed in an E. coli reporter strain. TMD-induced oligomerization causes dimerization of ToxR, activation of transcription and production of the reporter protein, -galactosidase.
