Amputation Induced Scrunching

Amputation-induced scrunching is a stereotyped, rapid whole-body contraction that planarians display after tissue removal, providing a visible behavioral response to injury. After amputation, wound-associated sensory signals activate coordinated contractions of the body-wall musculature, causing the animal to shorten and narrow repeatedly instead of maintaining its normal gliding movement. Researchers quantify scrunching frequency, duration, and onset to assess injury responses and examine how neural signaling, nociception, and regeneration interact. This behavior offers a simple, noninvasive assay for studying wound-induced physiology and testing how genetic or pharmacological manipulations alter responses during tissue repair.

Freshwater planarians exhibit three gaits (gliding, peristalsis, and scrunching) that are distinguishable by quantitative behavioral analysis. We describe a method to induce scrunching using various noxious stimuli, quantification thereof, and distinction from peristalsis and gliding. Using gene knockdown, we demonstrate the specificity of scrunching as a quantitative phenotypic readout.
