Antibody Compatibility

Antibody compatibility is the ability of antibodies, biological targets, and experimental reagents to work together without unwanted interference, a critical consideration in reliable immunological assays. Compatibility depends on factors such as antibody host species, isotype, target specificity, affinity, and the detection system, while cross-reactivity can produce background signal or misleading results. Researchers assess these factors when selecting primary and secondary antibodies for techniques including immunofluorescence, Western blotting, immunohistochemistry, and multiplex assays. Careful matching improves signal-to-noise ratios, preserves target discrimination, and supports accurate interpretation of protein localization, expression, and interactions in biological research.

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.
