Antigen Immunogenicity

Antigen immunogenicity is the capacity of an antigen to trigger a specific adaptive immune response, making it a central concept in biology, immunology, and vaccine development. It depends on how antigen structure, size, molecular complexity, dose, and delivery influence processing by antigen-presenting cells, presentation on major histocompatibility complex molecules, and activation of B and T lymphocytes; innate immune signals and co-stimulation also shape the response. Understanding these factors helps researchers design effective vaccines, evaluate antibody and cellular immunity, improve protein therapeutics, and predict why some antigens generate durable protection while others produce weak or limited responses.

Using a Sandwich ELISA to Determine the Immunogenic Glycoprotein Content in a Vaccine

2025

This video showcases an indirect enzyme-linked immunosorbent assay or ELISA sandwich immunocapture assay designed to assess immunogenic glycoprotein levels in vaccines. The assay plate, containing glycoprotein-specific antibodies, is incubated with serially diluted vaccine samples, including reference and test vaccines, followed by immunoassay to quantify glycoprotein content in the test vaccine.

A Technique to Assess the Immune Response against an Antigen by Inducing Skin Inflammation

2025

This video demonstrates an in vivo technique to assess a test antigen by inducing skin inflammation in a murine model. The test antigen—a hapten in an oil/acetone solution—is topically applied to the skin of the mouse ear, and inflammation of the skin is assessed after an adequate period.

In this antigen-driven colitis model, OT-II CD4+ T cells expressing a red fluorescent protein were adoptively transferred into RAG-/- mice that express a green fluorescent protein in mononuclear phagocytes (MPs). The hosts were challenged with Escherichia coli (E.coli) expressing the ovalbumin protein (OVA) fused to a cyan fluorescent protein (CFP).

Viral Antigen Microarray Assay to Detect Antibody Isotypes in Serum Against Viral Antigen
