Lymph Sac Formation

Lymph sac formation is an early step in embryonic lymphatic development in which specialized endothelial cells organize into fluid-filled lymphatic compartments, establishing the foundation for the lymphatic vessel network. During this process, venous endothelial cells acquire lymphatic identity through regulators such as Prox1, then migrate and sprout in response to lymphangiogenic signals including VEGF-C acting through VEGFR3; the resulting clusters expand, connect, and remodel. Studying lymph sac formation helps explain how lymphatic drainage, immune-cell trafficking, and tissue-fluid balance arise, while providing a developmental framework for investigating lymphedema, lymphatic malformations, and related disease.
