Auditory Bulla Dissection

Auditory bulla dissection is an anatomical technique used to expose the air-filled middle-ear cavity and adjacent structures for studying hearing biology. Typically performed under a microscope, the procedure removes surrounding temporal bone and opens the bulla to reveal the tympanic membrane, auditory ossicles, cochlear region, and associated tissues while preserving their spatial relationships. In biology research, this approach supports examination of ear development, middle-ear infection, trauma, ototoxicity, and hearing-related pathology. It also enables tissue collection, histological analysis, and evaluation of experimental treatments or devices, making it valuable for connecting ear anatomy with auditory function and disease mechanisms.

Auditory brainstem neurons of avians and mammals are specialized for fast neural encoding, a fundamental process for normal hearing functions. These neurons arise from distinct precursors of embryonic hindbrain. We present techniques utilizing electroporation to express genes in the hindbrain of chicken embryos to study gene function during auditory development.

The chicken auditory brainstem is comprised of nuclei responsible for binaural sound processing. A single coronal slice preparation maintains the entire circuitry while the cultured approach provides a unique preparation to study the development of neuronal structure and auditory function at the molecular, cellular and network levels.
