Automated Gel Extraction

Automated gel extraction is a laboratory technique that uses robotic instruments to isolate DNA fragments from agarose gel bands with minimal manual handling. An imaging system locates the desired band, after which the instrument excises the gel, dissolves its matrix, and purifies the DNA through silica-membrane or magnetic-bead binding, washing, and elution steps. In biology, this workflow supports cloning, sequencing, PCR product recovery, and other molecular applications requiring clean, size-selected nucleic acids. Automation can improve processing speed, consistency, sample traceability, and reproducibility while reducing contamination risks associated with repeated hands-on manipulation.

This manuscript describes an automated gel size selection approach for purifying DNA fragments for next-generation sequencing. The Ranger Technology provides complete automation of the entire process of agarose gel loading, electrophoretic analysis, and recovery of targeted DNA fragments allowing for high-throughput and high quality next-generation sequencing libraries.

TruTip is a simple nucleic acid extraction technology whereby a porous, monolithic binding matrix is inserted into a pipette tip. Consequently, the sample preparation format is compatible with most liquid handling instruments, and can be used for many medium to high-throughput clinical applications and sample types.

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column.
