Bhk-21 Cells

BHK-21 cells are a continuous, fibroblast-like cell line derived from baby hamster kidney tissue and widely used as a laboratory model in cell and molecular biology. In culture, these adherent cells attach to treated surfaces, proliferate in nutrient-supplemented medium, and can express introduced genes after transfection; their cellular machinery also supports replication of several viruses. Researchers use BHK-21 cells to study virus–host interactions, evaluate transfection systems, produce recombinant proteins, and develop vaccine-related processes. Their consistent growth and experimental versatility make them valuable for connecting cellular mechanisms with applications in virology, biotechnology, and biomedical research.

Here, we present a method to expand peripheral blood natural killer (PBNK), NK cells from liver tissues, and chimeric antigen receptor (CAR)-NK cells derived from peripheral blood mononuclear cells (PBMCs) or cord blood (CB). This protocol demonstrates the expansion of NK and CAR-NK cells using 221-mIL-21 feeder cells in addition to the optimized purity of expanded NK cells.

A method to generate human induced pluripotent stem cells (iPSCs) via retrovirus-mediated ectopic expression of OCT4, SOX2, KLF4 and MYC is described. A practical way to identify human iPSC colonies based on GFP expression is also discussed.

Ex Vivo Expansion of Natural Killer Cells from Peripheral Blood Mononuclear Cells

This video demonstrates a technique for the expansion of natural killer (NK) cells directly from peripheral blood mononuclear cells (PBMCs) using a feeder cell line. PBMCs are co-cultured with genetically modified lymphoblastoid feeder cells expressing membrane-bound interleukin-21 (mIL-21), and the culture is supplemented with the cytokines interleukin-2 (IL-2) and interleukin-15 (IL-15). Together, this cocktail of ligands leads to the prolonged expansion of NK cells with high purity.
