Blood Group Antibodies

Blood group antibodies are immune proteins that recognize specific antigens on the surface of red blood cells and are central to blood-group classification and transfusion safety. Produced after exposure to foreign red-cell antigens or present naturally in some blood-group systems, these antibodies bind selectively to target antigens and can trigger red-cell agglutination or complement-mediated destruction. Laboratory testing detects antibody–antigen reactions to determine blood type, identify unexpected antibodies, and assess donor–recipient compatibility. Understanding their specificity supports safe blood transfusion, investigation of hemolytic reactions, and evaluation of maternal–fetal incompatibility in pregnancy.

Prediction of Red Blood Cell Antibody Significance Using the Monocyte-Macrophage Assay

We have developed enhancements and updated methods for the existing monocyte-monolayer assay (MMA), in which macrophages are used to help better predict the clinical relevance of red cell alloantibodies in transfusion medicine and immunology. This assay is named the monocyte-macrophage assay (M-MA).

Methods for Quantitative Detection of Antibody-induced Complement Activation on Red Blood Cells

Here we describe two assays for measuring complement activation induced by antibodies against red blood cells. The major advantage over the current assays is their quantitative and easy-to-interpret nature.

A Semi-automated Approach to Preparing Antibody Cocktails for Immunophenotypic Analysis of Human Peripheral Blood

Whole blood Immunophenotyping is indispensable for monitoring the human immune system. However, the number of reagents required and the instability of specific fluorochromes in premixed cocktails necessitates daily reagent preparation. Here we show that semi-automated preparation of staining antibody cocktail helps establish reliable immunophenotyping by minimizing variability in reagent dispensing.

Human peripheral blood is commonly used for the assessment of the humoral immune response. Here, the methods for isolating human B cells from peripheral blood, differentiating human B cells into antibody (Ab)-secreting B cells (ASCs) in culture, and enumerating the total IgM- and IgG-ASCs via an ELISpot assay are described.
