Blood Platelets

Blood platelets are small, anucleate cell fragments in the circulation that preserve vascular integrity and limit blood loss after vessel injury. Produced from megakaryocytes in the bone marrow, they adhere to exposed collagen through von Willebrand factor, then activate, change shape, and release granule contents that recruit additional platelets and support coagulation; fibrin stabilizes the resulting plug. In biology and medicine, platelet function and count provide important indicators of hemostasis, inflammation, and disease. Studying platelets supports research into bleeding disorders, thrombosis, cardiovascular disease, transfusion practices, and therapies that regulate clot formation.

Purification and Activation of Platelets from Murine Whole Blood

In this video, we describe a protocol for purifying platelets from murine whole blood and their subsequent activation with the platelet agonist thrombin. When analyzed with flow cytometry, the purified resting platelets are characterized based on the expression of their cell-surface marker CD41, whereas purified activated platelets co-express both the surface markers CD41 and CD62P.

Platelet transfusion and hemostasis was modeled using blood reconstitution and microfluidic flow chambers to investigate the function of blood banking platelets. The data demonstrate the consequences of platelet storage lesion on hemostasis, in vitro.

Purification of Platelets from Mouse Blood

Here, mouse blood was collected in the presence of an anti-coagulant. The platelets were purified by iohexol gradient medium using low speed centrifugation. The platelets were activated with thrombin to investigate if they were viable. The quality of the purified platelets was analyzed by flow cytometry and microscopy.

A Microfluidic Flow Chamber Model for Platelet Transfusion and Hemostasis Measures Platelet Deposition and Fibrin Formation in Real-time

This paper describes an experimental model of hemostasis that simultaneously measures platelet function and coagulation. Platelet and fibrin fluorescence is measured in real-time, and platelet adhesion rate, coagulation rate, and onset of coagulation are determined. The model is used to determine platelet procoagulant properties under flow in concentrates for transfusion.
