Bone Marrow Stem Cells

Bone marrow stem cells are undifferentiated cells located in the marrow cavities of bones, where they maintain blood production and contribute to tissue repair. Hematopoietic stem cells self-renew and differentiate into red blood cells, white blood cells, and platelets in response to signals from the bone marrow niche, while mesenchymal stem cells can generate bone, cartilage, and other connective tissues. Their ability to replenish specialized cells makes them central to blood formation, immune function, and regenerative biology. Clinically, bone marrow stem cell transplantation treats disorders such as leukemia and some immune deficiencies, while research investigates their use in tissue engineering and disease modeling.

Mesenchymal stem cells are usually obtained from bone marrow and require expansion culture. When samples clot before processing, a protocol using the (enzymatic) thrombolytic drug urokinase can be applied to degrade the clot. Thus, cells are released and available for expansion culture. This protocol provides a rapid and inexpensive alternative to resampling.
