Brightfield Microscopy Analysis

Brightfield microscopy analysis is a light microscopy method that forms images from white light transmitted through a specimen, making it a foundational approach for examining biological structure and organization. The microscope focuses transmitted illumination through the sample, while differences in light absorption and scattering create contrast; staining can enhance otherwise transparent cells, tissues, or microorganisms. In biology, researchers use brightfield analysis to assess cell and tissue morphology, observe microbial growth, estimate cell abundance, and evaluate stained histological sections. Its accessible instrumentation and direct visual output support routine teaching, laboratory diagnostics, sample characterization, and quantitative image analysis.

Autophagy is a ubiquitous process that enables cells to degrade and recycle proteins and organelles. We apply advanced fluorescence microscopy to visualize and quantify the small, but essential, physical changes associated with the induction of autophagy, including the formation and distribution of autophagosomes and lysosomes, and their fusion into autolysosomes.

Capturing Tissue Repair in Zebrafish Larvae with Time-lapse Brightfield Stereomicroscopy

We present a protocol for capturing the dynamics of zebrafish larval tail fin regeneration on a whole-tissue scale using brightfield-based stereomicroscopy. This technique enables capturing the regeneration dynamics with single cell resolution. This methodology can be adapted to any stereomicroscope equipped with a CCD camera and time-lapse software.
