Bone Removal

Bone removal is the elimination of bone tissue from an organism, specimen, or anatomical preparation, and it is important for studying skeletal structure, tissue remodeling, and surgical intervention. In living organisms, specialized cells called osteoclasts dissolve the mineralized matrix and degrade collagen, while osteoblasts coordinate subsequent bone formation to maintain skeletal balance; in laboratory or clinical settings, mechanical or surgical methods may remove selected bone directly. These processes support investigations of bone development, fracture repair, disease, and regeneration, while controlled removal can provide access to underlying tissues or help treat damaged and abnormal bone.

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
