In Vitro Toxicity

In vitro toxicity is the study of harmful effects caused by chemicals, drugs, or other substances in biological systems maintained outside a living organism, such as cultured cells or tissues. In these tests, researchers expose cells to defined concentrations for controlled periods and measure responses including viability, membrane integrity, oxidative stress, or changes in cellular function. In vitro toxicity assays support early safety screening, environmental risk assessment, and investigation of mechanisms underlying tissue damage while reducing reliance on animal studies. Their controlled conditions enable rapid comparison of dose-dependent effects, although results require careful interpretation before predicting toxicity in whole organisms.

This protocol is designed to demonstrate exposure method of cell cultures to inhaled toxic chemicals. Exposure of differentiated air-liquid interface (ALI) cultures of airway epithelial cells provides a unique model of airway exposure to toxic gases such as chlorine. In this manuscript we describe effect of chlorine exposure on air-liquid interface cultures of epithelial cells and submerged culture of cardiomyocytes. In vitro exposure systems allow important mechanistic studies to evaluate...

2022

Drug-induced liver injury (DILI) is a major cause of drug failure. A protocol has been developed to accurately predict the DILI liability of a compound using a liver microphysiological system (MPS). The liver model uses the coculture of primary hepatic cells and translationally relevant endpoints to assess cellular responses to treatment.
