Brain Damage

Brain damage is the disruption or loss of normal brain structure and function caused by injury, disease, toxins, or reduced blood supply, and it can affect movement, sensation, cognition, and behavior. Damage may occur when physical trauma, stroke, infection, or oxygen deprivation injures neurons and supporting cells, triggering inflammation, cell death, and changes in neural signaling. In biology and medicine, studying brain damage helps researchers connect cellular processes with neurological symptoms and recovery. This knowledge supports the development of diagnostic methods, rehabilitation strategies, and treatments designed to limit secondary injury, protect neural tissue, and restore function.

Imaging Brain Damage Induced by Cerebral Hypoxia-Ischemia in a Mouse Model

2025

Source: Ouyang, Y., et al. Simultaneous PET/MRI Imaging During Mouse Cerebral Hypoxia-ischemia. J. Vis. Exp. (2015)This video demonstrates a technique to assess cerebral hypoxia injury via simultaneous positron-emission tomography (PET) and magnetic resonance imaging (MRI). An anesthetized mouse undergoes common carotid artery (CCA) ligation, reducing blood flow to one brain hemisphere and predisposing it to hypoxia-induced injury. PET and MRI imaging evaluate glucose metabolism and structural...

In this protocol, a novel application of scent discrimination is described. The Dig task is a reasonably inexpensive task that can be used to assess frontally-mediated cognition following brain damage.

Electron Microscopic Analysis of Axonal Damage in the Optic Nerve of Mice with Brain Injury

Using fluorescence microscopy, visualize the injured nerve, which exhibits a progressive increase in axonal swellings and disconnections.

Comet Assay for DNA Damage Detection in Cells

2025

The video demonstrates the comet assay for detecting DNA damage following exposure to harmful agents. Under alkaline conditions, the DNA unwinds and denatures, and upon electrophoresis, the undamaged and damaged DNA migrate differently in the agarose gel, forming a comet-shaped structure. The slowly migrating undamaged DNA forms the circular comet head, while the rapidly migrating damaged DNA fragments form the elongated comet tail.

Assessing DNA Damage Foci: To Identify Radiation-Induced DNA Damage Foci in Cancer Cell Line Using Immunofluorescence Staining

2023

This video describes a protocol to detect radiation-induced DNA repair foci in the colon cancer cell lines using immunofluorescence staining. DNA repair proteins get activated at the DNA damage site; therefore, we can detect them using specific antibodies and visualize them under a fluorescence microscope.
