Adipose Tissue Clearing

Adipose tissue clearing is a set of chemical and physical methods that makes fat-rich tissue transparent for three-dimensional imaging while preserving its cellular and molecular organization. These methods typically remove or solubilize lipids, reduce light scattering, and match the tissue’s refractive index with an imaging medium; some protocols also use permeabilization and fluorescent labeling to improve signal penetration. Cleared adipose tissue can then be examined by light-sheet or confocal microscopy to map adipocytes, blood vessels, immune cells, and neural structures. In biology, this approach supports studies of obesity, metabolism, inflammation, tissue remodeling, and interactions between adipose tissue and other organs.

Due to the high lipid content, adipose tissue has been challenging to visualize using traditional histological methods. Adipo-Clear is a tissue clearing technique that allows robust labeling and high-resolution volumetric fluorescent imaging of adipose tissue. Here, we describe the methods for sample preparation, pretreatment, staining, clearing, and mounting for imaging.

Here, we describe a simple, inexpensive and fast clearing method to resolve the 3D structure of both mouse and human white adipose tissue using a combination of markers to visualize vasculature, nuclei, immune cells, neurons, and lipid-droplet coat proteins by fluorescent imaging.
