Brain Tissue Preparation

Brain tissue preparation is the set of methods used to preserve, process, and section nervous tissue for microscopic examination and biological analysis. The workflow typically stabilizes tissue through fixation, removes water during dehydration, embeds it in a supportive medium, and produces thin sections for staining, imaging, or molecular assays. Careful control of these steps maintains cellular architecture, neural connections, and key molecular features while reducing tissue distortion. In biology, prepared brain samples support studies of development, neuroanatomy, disease pathology, and experimental treatment effects, enabling researchers to relate changes in cells and tissue structure to nervous system function.

We describe a protocol for transcardiac perfusion of mice, removal and sectioning of the brain, as well as immunoperoxidase staining, resin embedding, and ultrathin sectioning of the brain sections. Upon completion of these procedures, the immunostained material is ready for examination with transmission electron microscopy.
