Brood Pouch Dissection

Brood pouch dissection is a biological technique used to open and examine the specialized chamber that carries developing embryos or offspring, providing direct access to reproductive and developmental stages. During the procedure, the specimen is immobilized and dissected carefully under magnification so the pouch contents can be exposed, counted, measured, or assessed for developmental condition without obscuring delicate structures. This method supports studies of reproduction, embryology, and organismal development by revealing brood size, embryo morphology, and developmental timing. It can also help researchers evaluate how environmental conditions or experimental treatments influence reproductive success and offspring development.

Developing an Experimental Aneurysm Model in a Rabbit Using an Elastase-Treated Vessel Pouch

2025

Source: Wanderer, S., et al. Arterial Pouch Microsurgical Bifurcation Aneurysm Model in the Rabbit. J. Vis. Exp. (2020)This video demonstrates the creation of an experimental aneurysm model in a rabbit using an elastase-treated vessel pouch. After excising a segment of the right CCA, the pouch is treated with elastase, then sutured to the left CCA to restore blood flow. The resulting stress on the weakened pouch walls induces aneurysm formation.

An optimized technique for the microsurgical creation of arterial bifurcation aneurysms mimicking bifurcation human cerebral aneurysms is described. A venous pouch is sutured into an artificially created true bifurcation of both common carotid arteries. Facilitated microsurgical techniques and aggressive postoperative anticoagulation and analgesia lead to minimized morbidity rates and high aneurysm patency rates.

This method enables decellularization of a complex solid organ using a simple protocol based on osmotic shock and perfusion of ionic detergent with minimal organ matrix disruption. It comprises a novel decellularization technique for human hearts inside a pressurized pouch with real-time monitoring of flow dynamics and cellular debris outflow.
