Calcium Green Dye

Calcium Green dye is a fluorescent calcium indicator used to visualize changes in intracellular calcium concentration, an important signal in many biological processes. The dye contains calcium-binding groups that alter its fluorescence when they bind Ca2+, allowing changes in signal intensity to reflect calcium dynamics under appropriate imaging conditions. Researchers use Calcium Green dyes with fluorescence microscopy and related assays to study cell signaling, neuronal activity, muscle function, and calcium transport. These measurements help reveal how cells respond to stimuli and coordinate processes such as secretion, contraction, and communication.

Assessing T-cell Receptor-Induced Calcium Influx Using a Calcium Indicator Dye

2025

This video analyzes T-cell receptor-induced calcium influx in T-cells. Anti-CD3 antibodies activate the T-cell signaling pathways, leading to an increase in intracellular calcium ions that bind with Indo-1 dye. In flow cytometry, a spectrum shift from the calcium-free state to the calcium-bound state correlates with T-cell receptor-induced calcium influx.

Targeted-esterase induced dye loading (TED) supports the analysis of intracellular calcium store dynamics by fluorescence imaging. The method bases on targeting of a recombinant Carboxylesterase to the endoplasmic reticulum (ER), where it improves the local unmasking of synthetic low-affinity Ca2+ indicator dyes in the ER lumen.
