Metabolic Pathway

A metabolic pathway is a linked series of enzyme-catalyzed reactions that converts molecules within a cell, enabling organisms to obtain energy, build cellular components, and maintain life. In these pathways, the product of one reaction becomes the substrate for the next, while enzymes control reaction rates and regulatory signals adjust pathway activity to cellular conditions. Catabolic pathways such as glycolysis release energy, often capturing it as ATP or reducing equivalents, whereas anabolic pathways use energy to synthesize proteins, lipids, and nucleic acids. Mapping metabolic pathways helps researchers understand physiology, disease, drug responses, nutrition, and biotechnology.

13C-isotope labeling is a useful technique for determining the cell central metabolism for various types of microorganisms. After cells have been cultured with a specific labeled substrate, GC-MS measurement can reveal functional metabolic pathways based on unique labeling patterns in proteinogenic amino acids.

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.
