Cardiac Surface Imaging

Cardiac surface imaging is a set of methods used to visualize the heart’s outer surface and assess its structure, motion, and electrical or calcium activity. In optical approaches, fluorescent indicators on the epicardium change their emitted light as membrane voltage or intracellular calcium levels vary, while cameras capture these signals across the beating tissue. Spatial and temporal analysis of the resulting images can reveal conduction patterns, activation and recovery, and regional abnormalities. This information supports research on cardiac physiology, arrhythmia mechanisms, heart injury, and the evaluation of experimental therapies.

The sensitivity gain inherent to ultrahigh field magnetic resonance holds promise for high spatial resolution imaging of the heart. Here, we describe a protocol customized for functional cardiovascular magnetic resonance (CMR) at 7 Tesla using an advanced multi-channel radio-frequency coil, magnetic field shimming and a triggering concept.

High-speed Particle Image Velocimetry Near Surfaces

A procedure for studying transient flows near boundaries using high-resolution, high-speed particle image velocimetry (PIV) is described here. PIV is a non-intrusive measurement technique applicable to any optically accessible flow by optimizing several parameter constraints such as the image and recording properties, the laser sheet properties, and analysis algorithms.

Isolation and Genetic Manipulation of Adult Cardiac Myocytes for Confocal Imaging

Adult cardiac myocytes are primary cells that can be isolated from animal hearts and cultured for several days. Within this culture period adenoviral gene transfer can be used to express genetically encoded biosensors (GEBs) or fluorescent fusion proteins. Both approaches allow cellular investigations by means of confocal microscopy.
