Cell Culture Preparation

Cell culture preparation is the set of procedures used to establish, maintain, and ready living cells for controlled growth in vitro, providing a foundation for studying cell biology outside an organism. It typically involves selecting and handling a cell source, preparing sterile growth medium and culture vessels, introducing cells under appropriate environmental conditions, and monitoring attachment, viability, and proliferation. Careful preparation helps limit contamination and preserve relevant cellular characteristics. In biology, well-prepared cultures support experiments on cellular physiology, genetics, development, disease mechanisms, and responses to drugs or other treatments while improving reproducibility across research and teaching laboratories.

Chromosomes can be isolated from live cells such as lymphocytes or skin fibroblasts, and from organisms including humans or mice. These chromosome preparations can be further utilized for routine G-banding and molecular cytogenetic procedures such as fluorescence in situ hybridization (FISH), comparative genomic hybridization (CGH), and spectral karyotyping (SKY).
