Cell Population Gating

Cell population gating is the process of selecting and separating distinct groups of cells within a heterogeneous sample, most commonly during flow cytometry analysis. Researchers define regions, or gates, on plots of light-scattering and fluorescence measurements to exclude debris and doublets, then identify cells according to characteristics such as size, granularity, marker expression, or viability. Sequential gating creates a reproducible analytical strategy for quantifying subpopulations, comparing cellular responses, and isolating subsets for further study. In biology, this method supports immunophenotyping, disease research, developmental studies, and evaluation of treatment effects while helping distinguish genuine biological variation from technical noise.

Studying Proteolysis of Cyclin B at the Single Cell Level in Whole Cell Populations

Metaphase to anaphase transition is triggered through anaphase-promoting complex (APC/C)-dependent ubiquitination and subsequent destruction of cyclin B. Here, we established a system which, following pulse-chase labeling, allows monitoring cyclin B proteolysis in entire cell populations and facilitates the detection of interference by the mitotic checkpoint.

Measuring Alloreactivity in a Mixed Population of T Cells

This video demonstrates a method for assessing alloreactivity in mixed T cell populations by observing interactions with alloantigen-carrying dendritic cells, leading to the formation of immunological synapses. Differential staining and quantification of synapse-containing doublets via flow cytometry reveal T cell alloreactivity.

For many scientific studies requiring a biological and chemical analysis of cell populations the cells must be in a high state of purity. Fluorescence activated cell sorting (FACS) is a superior method in which to obtain pure cell populations.

Side Population Assay: A Method to Isolate Side Population of Tumor Cells from Zebrafish

The following protocol describes a method for isolating the side population of T-cell Acute Lymphoblastic Leukemia (T-ALL) tumor cells, consisting of stem cells, from an adult Zebrafish. The side population lacks prominent cell markers and hence we use Hoechst 33342 dye efflux assay to isolate them.

Assaying Blood Cell Populations of the Drosophila melanogaster Larva

Drosophila blood cells, or hemocytes, cycle between resident sites and circulation. In the larva, resident (sessile) hemocytes localize to inductive microenvironments, the Hematopoietic Pockets, while circulating hemocytes move freely in the hemolymph. The goal of this protocol is the standardized isolation and quantification of these two, behaviorally distinct but interchanging, hemocyte populations.
