Cell Type Specific Analysis

Cell type specific analysis is the study of molecular, structural, and functional differences among distinct cell populations within a tissue or organism. It works by identifying or isolating defined cell types and measuring features such as gene expression, protein abundance, morphology, or cellular activity, often using markers, microscopy, sequencing, or flow-based methods. In biology, this approach reveals how specialized cells contribute to tissue organization, development, disease, and responses to environmental changes. Comparing cell types can also identify rare populations, clarify disease mechanisms, and support targeted therapies by linking cellular characteristics to specific biological functions.

Heterokaryon Technique for Analysis of Cell Type-specific Localization

A flexible and efficient method for the characterization of cell type-specific protein localization and nucleocytoplasmic shuttling is described. This heterokaryon approach uses fluorescently-labeled fusion proteins to image protein localizations after cell fusion. The protocol is amenable to steady-state localizations or more dynamic determinations based on live cell imaging.

A method for producing Arabidopsis leaf protoplasts that are compatible with fluorescence activated cell sorting (FACS), allowing for studies of specific cell populations. This method is compatible with any Arabidopsis line that expresses GFP in a subset of cells.

The molecular basis of spatial-specific phytochrome responses is being investigated using transgenic plants that exhibit tissue- and organ-specific phytochrome deficiencies. The isolation of specific cells exhibiting induced phytochrome chromophore depletion by Fluorescence-Activated Cell Sorting followed by microarray analyses is being utilized to identify genes involved in spatial-specific phytochrome responses.

An Assay for Cell Cycle Analysis of Antigen-Specific CD8+ T Cells

This video demonstrates an assay to analyze antigen-specific CD8+ T cells in different cell cycle stages. Fluorescently-labeled viable cells are selected using flow cytometry, and the cells in the different cell cycle stages are identified by their DNA content using antibodies against cell proliferation-associated nuclear protein and a nucleic acid stain.
