Chemoattractants

Chemoattractants are chemical signals that direct cells toward higher concentrations of a substance, making them essential for coordinated movement in biology. Cells detect chemoattractant gradients through surface receptors, which activate intracellular signaling pathways that reorganize the cytoskeleton, establish front and rear polarity, and guide migration toward the signal. This process, known as chemotaxis, helps immune cells reach sites of infection or injury and supports embryonic development, tissue repair, and nervous system organization. Understanding chemoattractant signaling also informs research on inflammation, cancer cell invasion, and therapeutic strategies that modify cellular recruitment.

We describe a method to construct devices for 3D culture and experimentation with cells and multicellular organoids. This device allows analysis of cellular responses to soluble signals in 3D microenvironments with defined chemoattractant gradients. Organoids are better than single cells at detection of weak noisy inputs.

Eggs and the extracellular coatings around eggs frequently release peptides, proteins and small molecules that communicate with sperm to guide them to the egg thereby promoting fertilization. Using frog sperm we describe and compare two classes of assays used to detect sperm chemoattraction – sperm accumulation assays and sperm tracking assays.
