Cytoplasmic Aaa-atpase

Cytoplasmic AAA ATPases are ATP-powered molecular machines that remodel, unfold, or transport proteins within the cell, making them central to protein quality control and cellular organization. Typically assembled as hexameric rings, they bind ATP and hydrolyze it to drive conformational changes that grip protein substrates and translocate them through a central pore. These enzymes cooperate with proteases such as the proteasome, disaggregate damaged protein complexes, and regulate the assembly or activity of multiprotein machines. Studying their structure, substrate recognition, and energy-coupling mechanisms helps explain cytoplasmic proteostasis and supports research into neurodegeneration, infection, and potential therapeutic strategies.

Activation of Apoptosis by Cytoplasmic Microinjection of Cytochrome c

In this protocol, we describe the direct cytoplasmic microinjection of cytochrome c protein into fibroblasts and primary sympathetic neurons. This technique allows for the introduction of cytochrome c protein into the cytoplasm of cells and mimics the release of cytochrome c from mitochondria, which occurs during apoptosis.
