Cisterna Magna Puncture

Cisterna magna puncture is a procedure that accesses the cerebrospinal fluid (CSF) space at the base of the brain, typically for sampling or experimental analysis. Using anatomical landmarks, a needle is advanced into the cisterna magna, a CSF-filled cavity located between the cerebellum and medulla, allowing fluid collection while minimizing direct contact with brain tissue. In biology research, this technique supports studies of neuroinflammation, infection, protein biomarkers, drug distribution, and central nervous system physiology, particularly in animal models. Analyzing the collected CSF can reveal changes in cellular, molecular, and biochemical signals associated with neurological function or disease.

Source: Joshi, K., et al. Stereotaxic Surgical Approach to Microinject the Caudal Brainstem and Upper Cervical Spinal Cord via the Cisterna Magna in Mice. J. Vis. Exp. (2022)This video demonstrates the cisterna magna stereotaxic approach for targeted delivery to the brainstem and upper spinal cord. By accessing the cerebrospinal fluid-filled cisterna magna, this approach enables precise injections into areas that are difficult to reach using standard stereotaxic methods.
