Clonal Sphere Formation

Clonal sphere formation is a cell culture assay that evaluates the ability of individual cells to survive, proliferate, and generate multicellular spheres under defined conditions. Researchers typically seed single cells at low density in nonadherent, often serum-free medium, where cells with self-renewal capacity divide and form free-floating clonal spheres rather than attaching to a surface. Sphere number, size, and ability to form secondary spheres provide quantitative indicators of proliferative and stem-like properties. In biology, this method supports the study of stem and progenitor cells, tumor heterogeneity, differentiation potential, and changes in cellular behavior after genetic or pharmacological manipulation.
