Collagenase Solution

Collagenase solution is a laboratory reagent containing enzymes that break down collagen, the structural protein that helps organize connective tissues. By cleaving peptide bonds within collagen fibers under controlled conditions, it loosens the extracellular matrix while supporting the release of embedded cells. In biology, researchers use collagenase solutions to dissociate tissues, isolate primary cells, prepare samples for culture, and examine cell behavior in a more physiologically relevant context. Concentration, exposure time, temperature, and tissue type must be optimized because excessive digestion can reduce cell viability or damage surface proteins, whereas insufficient digestion may produce incomplete tissue disruption.

Isolation of Human Hepatocytes by a Two-step Collagenase Perfusion Procedure

A modified two-step collagenase perfusion procedure for isolation of human hepatocytes is described. This method can also be applied to other mammalian livers. The isolated hepatocytes are available in high yield and viability, making them a suitable model for scientific research in areas such as liver regeneration, pharmacokinetics and toxicology.

Pancreatic islets, also called the Islets of Langerhans, are a cluster of endocrine cells which produces hormones for glucose regulation and other important biological functions. This protocol explains a method for isolating pancreatic islets by first perfusing collagenase via Ampulla of Vater to digest the exocrine pancreatic tissue and a simplified density gradient centrifugation to purify the islets from mice.

Clinically relevant animal models of intracerebral hemorrhage (ICH) are needed to extend our knowledge of hemorrhagic stroke and to examine novel therapeutic strategies. In this study, we describe and evaluate two ICH models that implement unilateral injections of either autologous whole blood or bacterial collagenase into the basal ganglia (corpus striatum) of mice.
