Colony Screening

Colony screening is a method for identifying microbial colonies that carry a desired genetic construct or display a particular phenotype, making it essential for isolating successful clones from a mixed population. After cells grow on solid media, researchers examine individual colonies using selection markers, replica plating, color-based reporters, colony PCR, or restriction analysis to distinguish candidates from non-target colonies. In biology, colony screening supports recombinant DNA experiments, bacterial transformation, and strain development by confirming insert presence, orientation, or expression before further culture and analysis. Efficient screening reduces unnecessary work and helps researchers obtain genetically characterized colonies for downstream studies.

In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.

ScanLag: High-throughput Quantification of Colony Growth and Lag Time

ScanLag is a high-throughput method for measuring the delay in growth, namely lag time, as well as the growth rate of colonies for thousands of cells in parallel. Screening using ScanLag enables the discrimination between long lag-time and slow growth at the level of a single variant.
