Complex Iv Activity

Complex IV activity refers to the function of cytochrome c oxidase, the terminal enzyme complex of the mitochondrial electron transport chain that enables aerobic energy production. It accepts electrons from cytochrome c and transfers them to molecular oxygen, reducing oxygen to water while pumping protons across the inner mitochondrial membrane. This proton gradient drives ATP synthase and supports cellular energy metabolism. Measuring Complex IV activity helps assess mitochondrial respiration, oxidative phosphorylation, and the effects of genetic mutations, toxins, or disease-related damage on bioenergetic function. These analyses are important in studies of metabolism, neurobiology, aging, and mitochondrial disorders.

Electron Transport Chain: Complex III and IV

2023

During the electron transport chain, electrons from NADH and FADH2 are first transferred to complexes I and II, respectively. These two complexes then transfer the electrons to ubiquinol, which carries them further to complex III. Complex III passes the electrons across the intermembrane space to Cyt c, which carries them further to complex IV. Complex IV donates electrons to oxygen and reduces it to water. As electrons pass through complexes I, III, and IV, the energy released aids the pumping...

Flow Cytometry-Based Analysis of Dendritic Cell Activation Using Immune Complexes

2025

This video demonstrates a protocol for assessing the activation of dendritic cells using immunoglobulin G (IgG)-coated tumor cells. Dendritic cells internalize the IgG-coated tumor cells, process the tumor antigens, and present them on the surface through MHC-II molecules, along with the co-expression of the costimulatory molecule CD86. These dendritic cells then undergo MHC-II and CD86-targeted immunostaining and flow cytometry analysis to identify their activation state.

When compared to the previous paper protocol, implementation of a computerized glucose management system results in a substantial increase in blood glucose concentration measurements within the target range. Using a computerized glucose management system to monitor blood glucose levels, decreases in severe hypoglycemia (<40 mg/dL), clinical hypoglycemia (<70 mg/dL), and hyperglycemia (>180 mg/dL) also can be observed.

Recapitulation of an Ion Channel IV Curve Using Frequency Components

There are technical obstacles to measuring current flux through multiple ion channels simultaneously, and later discerning what portion of the transmembrane current is due to each channel type. To address this need, this method presents a way to generate the IV curve of individual channel types using specific frequency components.
