Computer-controlled Perfusion

Computer-controlled perfusion is the automated circulation of fluids through biological tissues, organs, or engineered culture systems, enabling precise control of their chemical and physical environment. A computer regulates pumps, valves, sensors, and feedback loops to adjust flow rate, pressure, temperature, oxygenation, and nutrient delivery while removing waste. In biology, this approach supports organ preservation, tissue engineering, cell culture, and studies of vascular and tissue function under controlled conditions. By maintaining stable, programmable conditions and recording changes over time, computer-controlled perfusion improves experimental reproducibility and helps researchers evaluate how cells and tissues respond to physiological or disease-related environments.

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.

The porcine model of liver normothermic machine perfusion (NMP), described here, can be successfully used to study NMP as a preservation strategy, a tool for viability assessment, and a platform for organ repair. It holds a high translational value, however it is technically challenging and labor-intensive.
