Continuous Light Incubation

Continuous light incubation is a controlled biological growth condition in which cells, tissues, plants, or microorganisms receive uninterrupted illumination for a defined period. By maintaining a constant light environment, researchers regulate light exposure while controlling variables such as temperature, intensity, and duration, allowing them to assess how organisms respond when the usual light-dark cycle is removed. This approach supports studies of photosynthesis, growth, metabolism, and photoperiodic responses, and can help reveal effects of prolonged illumination on biological rhythms, development, and experimental culture performance. Its use in laboratory research improves control over environmental conditions and enables reproducible comparisons between continuous-light and light-dark treatments.

We describe the reliable generation of non-Gaussian states of traveling optical fields, including single-photon states and coherent state superpositions, using a conditional preparation method operated on the non-classical light emitted by optical parametric oscillators. Type-I and type-II phase-matched oscillators are considered and common procedures, such as the required frequency filtering or the high-efficiency quantum state characterization by homodyning, are detailed.

Responding for food or drug-paired cues increases over the course of a period of abstinence and this may relate to an increased susceptibility to relapse behaviors. Here we detail a procedure for evaluating this "incubation of craving" in rats that have self-administered sucrose.

A Technique for the Prolonged Incubation of Neuronal Tissue of the Retina

2025

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.

Once removed from the body, neuronal tissue is greatly affected by environmental conditions, leading to eventual degradation of the tissue after 6 - 8 h. Using a unique incubation method, which closely monitors and regulates the extracellular environment of the tissue, tissue viability can be significantly extended for >24 h.
