Cuticle

Cuticle is a nonliving protective covering that forms the outer surface of many plants and animals, helping separate living tissues from the environment. In plants, cutin and surface waxes create a hydrophobic barrier that limits water loss while allowing controlled gas exchange. In arthropods, a chitin- and protein-rich cuticle provides structural support and protection, but growth requires periodic molting and replacement of the outer layer. Studying cuticle structure and function supports research in plant water relations, insect development, ecology, agriculture, and pest management, while revealing how organisms adapt to environmental stress.

Aerial plant organs are protected by the cuticle, a supramolecular biopolyester-wax assembly. We present protocols to monitor selective removal of epi- and intracuticular waxes from tomato fruit cuticles on molecular and micro scales by solid-state NMR and atomic force microscopy, respectively, and to assess the cross-linking capacity of engineered cuticular biopolyesters.

Isolation and Compositional Analysis of Plant Cuticle Lipid Polyester Monomers

Lipid polyesters constitute the structural components of two cell wall modifications, the plant cuticle and suberin-containing diffusion barriers. In this video, we describe a method to depolymerize cutin from whole delipidated leaves. The method can be applied to investigating mutants compromised in either cutin or suberin biosynthesis.
