Flexible Bone Experiment

The Flexible Bone Experiment is a simple biology investigation that demonstrates how bone composition determines its hardness and flexibility. In the standard procedure, an animal bone is soaked in an acidic solution, which dissolves much of its mineral content, particularly calcium salts, while leaving the collagen-rich organic framework relatively intact. As a result, the treated bone becomes noticeably softer and more bendable than an untreated bone. This model helps students connect bone structure with mechanical function and understand the roles of mineralization, collagen, and calcium in skeletal tissue. It also provides a practical introduction to tissue composition and experimental observation.

We describe a protocol for isolation and purification of neutrophils from mouse bone marrow by density gradient centrifugation and for neutrophil labeling using CellTracker dyes. This represents a simple, fast, reproducible and economical method for obtaining large numbers of neutrophils for downstream functional studies or adoptive transfer and tracking experiments.

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
