Cystic Duct Obstruction

Cystic duct obstruction is the blockage of the channel that carries bile from the gallbladder, most commonly by a gallstone, and it disrupts normal biliary function. When the duct is blocked, bile cannot leave the gallbladder normally, causing distention, chemical irritation, and potentially inflammation known as acute cholecystitis. Patients may develop right upper abdominal pain, nausea, or pain after fatty meals, while persistent obstruction can promote infection and complications. Understanding this process helps explain gallbladder physiology and supports diagnosis through clinical assessment and imaging, especially ultrasound, as well as decisions about treatment such as stone removal or gallbladder surgery.

Here we demonstrate our protocol for isolation of basal and submucosal gland duct cells from mouse tracheas. We also demonstrate the method of injecting stem cells into the dorsal mouse fat pad to create an in vivo model of submucosal gland regeneration.
